Abstract

Objective To investigate the effects of interleukin1(IL1)on expression of plasminogen activator inhibitor1(PAI1) and fibronectin(FN) in cultured human mesangial cells. Methods5BZNorthern blot analysis was used to detect the expression of PAI1 mRNA and FN mRNA in human glomerular mesangial cells cultured in media with or without IL1(5ng/ml).The activity of PAI1 and the quantity of FN in culture supernatants were also determined by fibrin plate method and ELISA. Results IL1 significantly increased the expression of PAI1mRNA and FNmRNA in glomerular mesangial cells.The levels of PAI1 and FN in the supernatants were markedly higher when the mesangial cells were cultured in the presence of IL1. Conclusion IL1 can upregulate the expression of PAI1 and FN both in mRNA and protein level.IL1 might contribute to glomerular sclerosis by inhibiting the degradation of extracellular matrix and promoting the synthesis of it.
