Influence of L-glutamine on the expression of HSP70mRNA in liver undergoing ischemia/reperfusion

Abstract
ObjectiveTo investigate the influence of L-glutamine on the expression of HSP70mRNA in liver and its protective role against ischemia/reperfusion damage.MethodsOne hundred and twenty male Wistar rats were divided randomly to three groups(n=40): sham-operation group(group 1),saline group(group 2) and glutamine group(group 3).Rats in group 2 were pretreated with 4ml 0.9% saline intraperitonally twice per day for 5 days consecutively.In group 3,rats were pretreated with Gln dissolved in 4ml 0.9% saline intraperitoneally twice per day for 5 days consecutively.The rats of groups 2 and 3 underwent total hepatic inflow occlusion for 35min utes through the pringle′s method.Ten rats from each group were randomly chosen and killed before the initiation of occlusion and at 2hours,4hours,24hours after reperfusion respectively.The levels of MDA in liver tissue were measured.The levels of serum TNF-α were detected.The serum concentrations of ALT,AST,LDH were assayed on a standard biochemistry autoanalyser.The expression levels of HSP70mRNA in hepatic tissue were assessed by reverse transcription polymerase chain reaction.ResultsCompared with group 2,the levels of MDA,ALT,AST,LDH and TNF-α in group 3 decreased significantly at 2hours,4hours,24hours after reperfusion(P0.05).The levels of HSP70mRNA expression in liver from group 3 were higher than those from group 1 and group 2 at 2hours,4hours,24hours after reperfusion(P0.05).ConclusionGln presupplement could alleviate the ischemia/reperfusion injury of liver and inhibit the release of proinflammatory factors after the pringle′s method,which might be related with the role of Gln in promoting the expression of HSP70mRNA.
