Abstract
Objective:To explore the effect of L-glutamine(Gln) on hepatic injury following total hepatic inflow occlusion.Methods:Male wistar rats were assigned randomly to three groups(n=40):Group 1 served as sham-operation group.Group 2 served as control,rats were pretreated with 4ml 0.9% saline intraperitonally twice per day on 5 consecutive days.In group 3,rats were pretreated with Gln dissolved in 4ml 0.9% saline intraperitoneally twice per day on 5 consecutive days.The rats from group 2 and 3 underwent total hepatic inflow occlusion for 35 min through the pringle′s manoeuvre.Ten rats from each group were randomly chosen and killed before the initiation of occlusion and at 2 h,4 h,24 h after reperfusion respectively.The levels of MDA,SOD,GSH in liver tissue were measured.The levels of serum TNF-α and portal-vein endotoxin were detected.The serum concentrations of ALT,AST,LDH were assayed on a standard biochemistry autoanalyser.Results:Compared with group 2,the levels of GSH and SOD in liver from group 3 increased after reperfusion(P0.05),that of MDA in liver decreased(P0.05),that of ALT,AST,LDH,TNF-α and endotoxin from group 3 decreased(P0.05).Conclusions:Gln presupplement can alleviate the ischemia/reperfusion injury of liver after the pringle′s manoeuvre in some extent,which may be related with the role of Gln in reducing the translocation of gut-derived endotoxin and release of proinflammatory factors.
