Abstract

A PCR based detection method was developed using a pair of primers designed from a conserved region of the pseudorabies virus(PRV) gH gene. This method uses an improved DNA extraction approach to isolate DNA from PRV infected PK-15 cells and optimized PCR conditions to detect the presence of PRV sequence. A 355 bp fragment was amplified from PRV infected PK-15 cells, but not from PRRSV?¢PPV?¢HCV?¢JEV and PK-15 cells. This amplified fragment is specific for PRV DNA based on Nae¢?digestion pattern. The results showed that this improved PCR approach could be a fast, simple and specific detection method for PRV.
