Abstract
Objective: To express vWF binding domain of platelet GPⅠbα in Pichia pastoris. Methods: The gene of vWF binding domain of platelet GPⅠbα was obtained by polymerase chain reaction(PCR) from recombinant plasmid pcDNA3.1-GP Ⅰbα.The recombinant expression vector was constructed and was electroporated into pichia pastoris GS115. The transformants were screened by G418 and induced by methanol. The expressed protein was identified by SDS-PAGE and dot blot. Results: Recombinant plasmid pPIC9k-GP302 was obtained.Positive colonies were obtained by G418 screening and were induced by methanol.The expressed protein had ability to bind anti-GPⅠbα monoclonal antibody, which was identified by dot blot. Conclusion: vWF binding domain of platelet GPⅠbα was expressed in Pichia pastoris.
