Abstract

Objective: To establish a new assessment method for animal model of choroidal neovascularization(CNV).Methods: In this model,CNV was induced by laser photocoagulation in C57BL/6 mice with the krypton red laser.Animals from each group were sacrificed and were perfused through the heart with fluorescein-labeled dextran(FITC-Dextran) on day 1,3,5,and 7 post laser.Retinal pigment epithelium-choroid-scleral tissue, after using a mAb specific for elastin staining, was examined to determine the incidence and size of CNV complex by confocal microscopy. Results: Confocal micrograph of the neovascular complex in C57BL/6 mice showed the new vessels(green),and damaged Bruch's membrane(BM) and RPE were stained red for elastin.CNV appeared on day 3 post laser, reaching the peak on day 7 post laser. Conclusion: The new assessment method of animal model of choroidal neovasculation(CNV) is feasible, reliable and visualized.
