Abstract
Objective To explore the effect of different methods of antigen retrieval on the result of immunohistochemical staining of Ki-67.Methods Antigens were retrieved respectively by using trypsin,citric microwave,citric autoclave,Tris/EDTA microwave and Tris/EDTA autoclave in tumor paraffin sections from nude mouse,followed by immunohistochemical staining.Results After antigen retrieval with citric microwave,the positive staining intensity of nuclear protein Ki-67 was enhanced,and Ki-67 was better located by using of this method than the others.Conclusion In the immunohistochemical staining methods of Ki-67,citric microwave is the best one to retrieve antigen.
