Effect of recipient hypoxic preconditioning on transplanted liver via protecting intestinal barrier function

Abstract

Objective:To investigate the effect of recipient hypoxic preconditioning on trans-planted liver reperfusion injury.Methods:Male Sprague-Dawley(SD)rats were randomly divided into three groups,A:OLT group(n=20),B:hypoxic preconditioning-OLT group(n=20),C:shame operation group(S,n=12).Orthotopic rat liver transplantation was performed according to Kamada two-sleeve approach.Plasma endotoxin,liver function,SOD,MDA were detected two hours post operation.Liver tissue samples and ileal mucosal samples were studied by electronic-microscopy and optic-microscopy.Twelve recipients were used to observe post-operation survival rate.Re-sults:The ALT,AST,TBIL and MDA levels in groups A were higher than those in groups B while SOD activity changed reversely.Endotoxin level of B group was significantly lower than that of group A.One week survival rate in group B was significantly higher than that in group A.There was no sig-nificant differences in cell morphology among the groups.Conclusion:Recipient hypoxic precon-ditioning may protect intestinal barrier function and relieve reperfusion injury of the liver graft.
