Abstract
β-glucosidase from rubber seed was immobilized on chitosan microsphere crosslinked with glutaraldehyde.Based on single factor experiments,taking chitosan concentration,acetic acid concentration and ratio of NaOH solution and ethanol as independent variables,activity recovery as response value,response surface methodology(RSM) was applied to optimize the method of immobilization.The optimal manufacturing conditions of microsphere were:2.5% chitosan,2% acetic acid,7.5% NaOH solution,7.5% NaOH solution:95% ethanol(v/v)=1:1,let stand for 14 h.The enzyme activity recovery of immobilized β-glucosidase was 74.97%.
