Effect of matrine on the expression of CC10 mRNA in lung adenocarcinoma A549 cell line

Abstract

Objective To observe the effects of matrine on the cell growth, ap optosis and CC10 mRNA expression in lung adenocarcinoma A549 cell line. Methods Human lung adenocarcinoma A549 cells were cultured for 72 hours and divided into experimental group A, B, C, D and E based on the concentrations of matrine rang ed from low to high and one control group. The inhibitory action of matrine on t he growth of lung adenocarcinoma A549 cell line was measured by MTT, the effect of matrine on apoptotic morphology was observed by Hoechst33258 fluorochrome sta ining, the apoptotic rate was evaluated by AnnexinV-PI, and the expression of C C10 mRNA was detected by RT-PCR. Results Matrine with different concentrations could inhibit the cell growth and induce apoptosis in lung adenocarcinoma A549 c ell line. The inhibitory rate of cell growth and apoptotic rate in all experimen tal and control groups were 24.7%and 5.12%, 33.1%and 8.36%, 44.3%and 10.81 %, 61.7%and 17.10%, 74.2%and 25.67%, 6.2%and 3.06%, respectively. There w as a significant difference among the experimental groups and the control group( P 0.05). The positive rates of CC10mRNA expression in five experimental groups were 1.03±0.07, 0.87±0.03, 0.74±0.04, 0.62±0.06 and 0.49±0.04, respectively . Conclusion The effects of matrine on inhibiting the cell growth and inducing a poptosis in A549 lung adenocarcinoma cell line may be associated with the up-re gulation of CC10 mRNA expression.
