Protective effect of total ginkgolides on cerebral ischemia in mice and rats

Abstract

AIM To investigate the protective effects of total ginkgolides (TG) on anoxia and acute cerebral ischemia in mice and focal cerebral ischemia in rats. METHODS Anoxia was produced by close hypoxia; Acute cerebral ischmia was produced by the occlusion of bilateral common carotid arteries with vagus nerves of mice; Focal cerebral ischemia was produced by permanent occlusion of the proximal of the left middle cerebral artery (MCA). The infarct area was measured by 2,3,5 triphenyltetrazolium chloride (TTC) staining technique. The extent of neurological deficits was observed. RESULTS TG (32 mg·kg -1 , ip×8 d) significantly prolonged the survival time of mice subjected to acute cerebral ischemia by occlusion of bilateral carotid arteries and decreased the death rate. TG (4, 8, 16 mg·kg ·1 , ip×8 d) also markedly decreased the infarct size and ameliorated neurologic deficits score of rats subjected to MCAO. CONCLUSION TG has protective effects on cerebral ischemia.
