Comparison of the proliferative effects of porcine pseudorabies virus strain SD-1 in several different cell cultures

Abstract

To select the most suitable cell line for the proliferation of porcine pseudorabies virus SD-1 strain,taking BHK-21,PK-15,CEF and Vero cells as the vaccination target of the strain,the proliferative effects of porcine pseudorabies virus SD-1 strain were compared in BHK-21,PK-15,CEF and Vero cell cultures.The cytopathic time and characteristics were observed,and the virus titers(TCID50) were measured respectively,and the neutralization test was used to the vaccinated groups with PRV-positive serum(titer 1∶ 128).The results showed that the pseudorabies virus SD-1 strain could all produce significant cytopathic effect(CPE) in BHK-21,PK-15,CEF and Vero cells,CPE began to appear first at 12 hours,15 hours,20 hours,and 24 hours after inoculation,respectively.The TCID50 in the four kinds of cell cultures were 10-7.3/0.1 mL,10-6.3/0.1 mL,10-6.7/0.1 mL,and 10-5.7/0.1 mL,respectively.And there was no CPE in the neutralization groups.Therefore,it is suggested that BHK-21 cell can be used as the cell line for the proliferation of porcine pseudorabies virus SD-1 strain.
