Effect of cisplatin on apoptosis of cervical cancer cell line HeLa and the expression of FHIT,Ki67 and Bax proteins

Abstract

Objective To investigate the apoptosis of HeLa cell,the expression of FHIT,Bax and Ki67 proteins induced by cisplatin,and to explore its mechanism.Methods The effect of cisplatin on growth of HeLa cell was analyzed by MTT assay.The apoptosis was measured with cytometry after treated with cisplatin.Immunohistochemical analysis was performed to detect the expression of FHIT,Ki67 and Bax proteins.Results Cisplatin inhibited proliferation of HeLa cell in a time-and dose-dependant manner.The apoptosis rate significantly increased after 24 h treatment with different concentrations of cisplatin by flow cytometry.The expression of FHIT and Bax proteins increased gradually after treatment with cisplatin in HeLa cells(P0.05),while the expression of Ki67 protein decreased significantly(P0.05).Conclusion Cisplatin can inhibit the growth of HeLa cell and induce apoptosis by up-regulating the expression of FHIT and Bax proteins and down-regulating the expression of Ki67.
