Effect of Celecoxib on Chemotherapy Sensitization for Cervical Cancer Cells

Abstract

Objective To investigate the effect and its mechanism of celecoxib on cisplatin-induced proliferation and apoptosis in cervical cancer Hela cells.Methods The proliferation of celecoxib on cisplatin-induced cervical cancer Hela cells was examined by MTT assay.The apoptosis of Hela cells were tested by PI staining flow cytometry.The expression and their activity change of Bcl-2,Bax protein in Hela cells were investigated by Western blot.Results 5,10μmol/L concentrations of celecoxib combined with cisplatin significantly increased the cytotoxicity of cisplatin to Hela cells.Celecoxib(5μmol /L) significantly promoted cisplatin to induced Hela cells apoptosis.the expression levels of Bcl-2 protein were decreased and the expression level of Bax protein was increased in Hela cells treated with celecoxib in a concentration dependent manner(P0.05).Conclusion Celecoxib could enhance the effect of growth inhibtion and apoptosis induction of cisplatin on Hela cells.The effect of chemosensitization to cisplatin of celecoxib on Hela cells may be associated with downregulating the expression of Bcl-2 protein and upregulating the expression of Bax protein.
