Abstract
Objective To explore the effects of pirarubicin(THP) on the apoptosis and the activity of nuclear factor kappa B p65(NF-κB p65) of HL-60 cells.Methods The experiment was carried in two parts.In the first part,after being cultured in RPMI 1640 for 2 h,the HL-60 cells were divided into A and B groups to continue the culture for another 12 h,with PBS added in the media of group A,and THP 1,10 and 100 μmol/L added in the media of the respective B subgroups.The apoptosis of HL-60 cells was determined by DNA ladder electrophoresis and flow cytometry(FCM).In the second part,The cells were grouped and cultured in the way similar to that in the first part,except for that the cultures were continued for another 3 h before the activation of NF-κB p65 of HL-60 cells was determined by FCM.Results The rates of apoptosis of HL-60 cells induced by THP 1,10 and 100 μmol/L were(31.78±4.31)%,(47.25±5.27)% and(56.49±1.59)%,respectively,higher than the control(6.46±1.45)%,P0.05;and the rates of activation of NF-κB p65 were(16.21±1.20)%,(23.98±3.21)% and(32.44±2.89)%,respectively,higher than the control(8.44±2.20)%,P0.05.Conclusion THP increases the apoptosis,and at the same time,promotes the activation of NF-κB p65 of HL-60 cells in dose-dependent manner.
