Abstract
Objective:The study is to find out the inhibition action of TNF-related apoptosis-inducing ligand or APO-2ligand (TRAIL)on ovarian carcinoma cell line cultured in vitro.Methods:RT-PCR was used to assay the expression of TRAIL receptors of DcR1、DcR2、DR4、DR5and4,5-dimethyl thiazole(MTT)was applied to assay the inhibiting action of various concentration of TRAIL on different ovarian carcinoma cell line of3AO,SKOV 3 and OVCAR 3 ,with Cisplatin(cDDP)as the con-trol.The apoptotic rates and the changes of cell cycle were measured by flow cytometry(FCM)and morphological changes in cell line3AO with the existence of TRAIL were compared with scanning electron microscope(SEM)and Hematoxylin-eosin(HE)staining.Results:①The growth of human o-varian carcinoma cell line3AO,SKOV 3 and OVCAR 3 was effectively inhibited by TRAIL with time-ef-fect and concentration effect characteristics,(P0.05);②Treated with different concentration of TRAIL,the cell line3AO reached cell apoptosis peak of subdiploid before the G1phase of cell cycle and assumed typical cell apoptosis configuration under the action of TRAIL in contrast to the necrosis configuration under cDDP.Conclusions:①TRAIL can effectively inhibit the growth of ovarian carcino-ma cell line with time-effect and dose-effect characteristics.②TRAIL can induce the apoptosis of cells in different ways.
