Abstract

Objective To establish DDP-resistant human NSCLC cell line(A549 /DDP)and to study the relationship between drug-resistant cell and Side population Cell(SP cell).Methods Establishment of DDPresistant human NSCLC cell line A549 /DDP using high-does and interval-inactivation methods.The characteristic of drug-resistant cell was detected by MTT assay.The percentage of SP cell was determined by Hoechst33342 staining and Flow Cytometry(FCM)assay.Results A549 /DDP cell line was established after 7-month treatment,which could resist to DDP with resistance index of 5.957.A549 /DDP cell exhibited cross resistance to many other chemotherapeutic agents.The percentage of SP cells was about 1.09 % in A549 cell line and 0.31% in A549 /DDP cell line.Conclusion A549 /DDP cell line shows the typical multidrug-resistant phenotype.The percentage of SP cells is similar between A549 and A549 /DDP.
