Effects of emodin on serum leptin of rats with severe acute pancreatitis

Objective To observe the protective effecs of emodin on the alteration of serum leptin in severe acute panereatitis(SAP)rats.Method A total of 72 Wistar rats were randomly divided into 3 groups:sham- operation group(SO group,n=12),SAP group(n=12),and emodin group(EM group,n=12).Rats of SAP group were induced by intraduetal injection of 6% sodium taurocholate(1.0 ml/kg)in SAP group.In EM group,the model of SAP was reproduced first and 1hen emodin was injected into abdominal cavity(1 ml/kg, twice/d),and saline was injected instead of emodin in SAP group.In SO group,rats were only intraductal ly injected saline instead of taurneholate.Serum levels of amylase,lipase,TNF-α,1L-10 and leptin levels were determined.The pathologic changes of pancreas were observed after 24 hours and 48 hours.The immunohistochemistry method(SP method)was used to examine the expression of leptin.Results At 24 hours after SAP,serum level of amylase was higher in SAP group(9784.800±2634.894)U/L than in SO group (611.33±209.75)U/L and EM group(7832.67±2641.29)U/L(P0.01).At 24 hours after SAP,serum level of TNF-αwas higher in SAP group(517.76±9B.98)pg/ml than in SO group(249.08±83.02)pg/ml and EM group(259.10±50.08)pg/ml(P0.01).At 24 hours after SAP,serum level of IL-10 in EM group was (677.991±154.067)pg/ml,higher than SAP group(637.73±323.90)pg/ml and SO group(339.88±104.71)pg/ml.The leptin level of SAP group and EM group were increased,and 24 hours later,the leptin level in EM group was(15.91±4.61)ng/ml,which was higher than in SAP group(13.61±8.75)ng/ml(P0.05).In SAP group,rats had severer interstitial edema,hemorrhage,necrosis and neutrophil infiltration than in EM group.In EM group,the pathologic changes of pancreas were less severe,and the positive rate of leptin expression was markedly higher than that in SO group and SAP group.Conclusions In the early stage of SAP, emodin can increase leptin level,inhibits the inflammatory mediates and decreases the pancreas injury.
