Effect of ginkgolide B on plasma level of tumor necrosis factor alpha in severe acute pancreatitis rat model

Abstract

Objective To observe the plasma level of tumor necrosis factor alpha(TNFα)in severe acute pancreatitis(SAP) rat model and the efficacy of ginkgolide B(BN52021)in treating SAP.Methods Wistar rats were randomly assigned to model group(SAP,n=15),treatment group(BN,n=15),and negative control group(NC,n=15).SAP was induced by retrograde infusion of 5% sodium taurocholate into the pancreatic duct of Wistar rats.SAP group received the femoral vein injection of isotonic sodium chloride 0.5 ml/100mg 15 minutes after induction of SAP;BN group received BN52021(5 mg/kg),while NC group received only abdominal incision.At 1,6 and 12 hours after operation,rats were sacrificed for determination of plasma TNFα by ELISA.Results At 1 h post-operation,there was no significant difference in the plasma TNFα levels between SAP group(420.67±262.22 pg/ml),BN group(403.88±177.92 pg/ml),and NC group(369.62±39.73 pg/ml)(P0.05).At 6 h and 12 h post-operation,the plasma levels of TNFα in SAP group(856.33±207.25 pg/ml and 961.56±(415.49) pg/ml,respectively) were significantly higher than those in NC group(369.14±16.28 pg/ml and 416.43±26.54 pg/ml,respectively)(P0.05) and BN group(415.15±88.08 pg/ml and 692.06±69.31 pg/ml,respectively)(P0.05).Conclusions The plasma level of TNFα in SAP group is significantly increased in SAP and that in SAP rats treated with ginkgolide B is significantly decreased.
