Abstract
【Objective】To study the expression of endostatin after EJ cells being infected by rAAV-Endostatin and the inhibition effect on human umbilicus vein endothelial cell(HUVEC) chemotactic movement,make ready for antiangiogenesis gene therapy on bladder cancer.【Methods】EJ cells were infected with rAAV-Endostatin,the supernatant was collected 24 h later and endostatin concentration within it was assayed by ELISA;the inhibition effect of the supernatant on HUVEC chemotactic movement was assayed by Transwell test too.【Results】Endostatin concentration in the supernatant was 54.09 ng/ml,the inhibition rate on HUVEC chemotactic movement was 37.45% in Transwell test.【Conclusion】rAAV-Endostatin is an effective vector of endostatin gene therapy,and thus makes a foundation of endostatin gene therapy for bladder cancer.
