Abstract
Objective To optimize in vitro culture conditions of Blastocystis hominis in RPMI1640 medium. Method The acidity, inoculation-dose, sorts and concentration of serum, temperature, oxygen, and selection of stool specimens were studied. Result pH 7.5,2×105 cell/tube,20% bovine serum,1×105 IU penicillin and 1.6×105 IU streptomycin,37℃ are the best in vitro culture conditions for Blastocystis hominis. Conclusion Mucous and bloody stool specimens that contained mainly granular forms is a better source of Blastocystis hominis for the culture. The optimal condition for culturing Blastocystis hominis is in RPMI1640 at pH7.5 with 20% calf serum, streptomycin, and penicillin, under anaerobic condition at 37℃. Blastocystis hominis can culture for long time with subculture at every 6 days.
