Abstract
Objective:To study neural stem cell(NSCs) transplantation to promote axonal regeneration and the function of hind limbs in rat after spinal cord injury.Methods:NSCs in logarithmic phage were labeled with BrdU.Spinal cord injury(SCI) model of Sprague Dawley rats was generated with electrocircuit control spinal cord injuring device.Thirty SD rats were randomly divided into groups sham,SCI,and NSC.The NSCs were transplanted into the injured site three days after SCI.Then the NSCs labeled with Brdu and NF200 were detected by immunohistochemisty,Hind limb function of the rats was evaluated by an independent observer who is blind to the grouping using BBB open field locomotion rating score.Results:NSCs can be detected in the spinal cord after transplantation.The mean OD of NF200 was higher than that in SCI group(P0.05).Twenty-eight days after transplantation,the number of NF200 immunostaining positive axons was increased remarkably than that in the SCI group(P0.01),and the BBB score of NSC group was significantly higher than that in the SCI group(P0.01).Conclusion:Transplantation of NSCs can survive in the injured site of spinal cord and promote the axonal reconstruction,and the function of the hind limb will be improved significantly in rats after SCI.
