Effects of Flavonoid on the Proliferation of Prostate Cancer Cells PC-3

Abstract

To explore the feasibility of dietary intervention through resesrch on effects of flavonoid on the proliferation of prostate cancer cells (PC 3). Prostate cancer cells were grown in RPMI 1640 medium supplemented with 10% bovine serum and 5 ml 10 000 U/ml of penicillin/streptomycin in an incubator maintained at 5% CO 2 and 37℃. The respective test compound was added into fresh medium,the control cells received only the vehicle(dimethyl sulfoxide,DMSO).The proliferation of PC 3 cells was analyzed by MTT assay, 3H TdR incorporation assay and flow cytometry. Compared with the DMSO control,this study demonstrated that the proliferation and DNA synthesis of prostate cancer cells treated with genistein(25 mmol/L,72 h),daidzein,(75 mmol/L,72 h) and glycitein (75 mmol/L,72 h) respectively were markedly inhibited.The results indicated that there is a time dependent and dose dependent relationship. [Conclusion] Soy isoflavones (genistein,daidzein,glycitein) could significantly inhibited the proliferation of PC 3 cells.The findings give a support to the hypothesis that dietary isoflavones can prevent the incidence of prostate cancer.
