Influence of IGF-1 on expression of BMP-2 and BMP-7 genes in osteoblast

Abstract

Objective To study the influence of IGF-1 on the expression of BMP-2 and BMP-7 genes in osteoblast. Methods Cultured rat osteoblasts were treated with IGF-1 in different concentrations. Total RNA was exfracted the from the cells. BMP-2 and BMP-7 cDNA were amplified by RT-PCR. β-Actin cDNA was also amplified at the same time and used as control. The pictures of PCR production was scanned, The specific values of light-density of BMP-2/β-actin and BMP-7/β-actin were calculated, and the expression levels of BMP-2 and BMP-7 genes were determined. Results IGF-1 increased the expressions of BMP-2 and BMP-7 genes at the level of transcription dose-dependently. Conclusion IGF-1 induces the proliferation of osteoblast probably by increasing the expressions of BMP-2 and BMP-7 genes.
