Abstract

Objective:To evaluate the specificity of three-color flow cytometry in adult acute leukemia immunophenotyping.Methods:Immunophenotyping of one hundred and ninety seven adults with acute leukemia was performed by three-color flow cytometry analysis using CD45/SSC gating.Results:Lymphoblastic antigens in all acute lymphoblastic leukemia cases were expressed,in which CD13 and CD33 were 46.7% and 11.1%,respectively;myeloid antigens in all acute myeloid leukemia cases were expressed,and CD34 was expressed in only one of M3 and no expression of HLA-DR was found in all of M3;CD7 and CD34 expression were twelve and twenty one in AML,respectively;Among one hundred and ninety seven cases of AL,5.6% was HAL, six cases of which were B/M.Conclusion:Gating of CD45/SSC can eliminated effect of normal cells on blasts in bone marrow,immunophenotyping results were useful to subtyping and diagnosis of acute leukemia.
