The Myocardial Apoptosis and the Expression of Fas and FasL Genes in Ischemic Myocardium of Rats

Abstract

Objective To study the myocardial apoptosis and the expression of Fas and FasL genes in rats with ischemia-injured myocardium induced by isoprenaline(ISO).Methods Adult healthy SD rats were randomly divided into control group and myocardial ischemic injury group(injury group).The rat myocardial ischemia-injured model was induced by injecting large dose ISO intraperitoneally.Terminal deoxynucleotidyl transferase-mediated dUTP nickend labeling(TUNEL) was used to detect the myocardium apoptosis.Western blotting and immunohistochemistry were used to assay the expression of Fas and FasL proteins quantitied by HPIAS10009 computerized image analytical instrument.Results Compared with the control group,apoptotic index of cardiomyocytes increased in injury group(P0.01),the expression of Fas and FasL genes in rat cardiomycytes of injury group was significantly higher than that of control group(P0.01).Conclusion Myocardial apoptosis and the overexpression of Fas and FasL genes suggest that apoptosis and Fas/FasL system are involved in the occurrence and development of myocardium ischemia-injury in rats.
