Protection of ligustrazine to the mucosa of small intestine on acute necrotizing pancreatitis in rats

Abstract

Objective: To evaluate the role of microcirculatory disorder(MCD),and study the therapeutic effectiveness of ligustrazine to the mucosa of intestine on acute necrotizing pancreatitis in rats.Methods: A total of 192 SD rats were divided into three groups at random(each with 64 rats): control group(C);pancreatitis(P) group;ligustrazine treatment(T) group.Radioactive biomicrosphere technique(RMT) was used to measure the intestinal blood flow at 0.5h、2h、6h and 12h after the induction of ANP.The samples of pancreas,distal ileum were collected to observe pathological changes using a validated histology score.Intestinal tissues were also used for examination of myeloperoxidase(MPO) and PMN. Results: Comparison of P group and C group,the blood flow in P group was significantly lower than that of C group(P0.01);the pathological changes and infiltrating degrees of neutrophils were aggravated significantly in P group.The intestinal MPO activity in P group were significantly higher than those in C group(P0.01).Comparison of T group and P group,the blood flow of intestine in T group was significantly higher than that of P group after 2h(P0.01);the pathological changes and infiltrating degrees of neutrophils were alleviated significantly in T group.MPO activity were significantly lower than those in P group(P0.01 or P0.05).There were significant negative correlation between the blood flow of distal ileum and myeloperoxidase(MPO) activity and the pathological changes of intestinal tissues in P group. Conclusion: MCD was an important factor of intestinal injury in ANP.Ligustrazine can ameliorate the condition of MCD and alleviate the damage of pancreas and intestine.
