Abstract
OBJECTIVE To evaluate the role of microcirculatory disorder(MCD) in acute necrotizing pancreatitis (ANP) in SD rats,and evaluate the therapeutic effectiveness of ligustrazine on lung injury in ANP.METHODS A total of 192 SD rats were randomly divided into three groups:normal control group (C),ANP group without treatment(P) and ANP group treated with ligustrazine(T). The ANP model was induced by injection of 5% sodium taurocholate under the pancreatic membrane (4 ml·kg~-1). Normal control group was received isovolumetric injections of 0.9% physiological saline solution using the same method. T group was injected ligustrazine(10 ml·kg~-1) via portal vein. Radioactive biomicrosphere technique(RMT) was used to measure the blood flow at 0.5,2.0,6.0,12.0 hour after the induction of ANP.Eight rats in each group was only used for blood flow determinations by the intravenous injections of RMT. The samples of pancreas ,lung were collected to observe pathological changes using a validated histology score. Lung tissues were also used for examination of myeloperoxidase (MPO) activity and dry/wet ratio. RESULTS Comparing ANP group with normal control group, the blood flow in ANP group was significantly lower than that of control group (P0.05).The pathological changes were aggravated significantly in SAP group.Longer the time was,more severe the lung injuried.The tissue injury was closely correlated with blood flow of lung and wet/dry ratio.MPO activity in ANP group were significantly higher than those in control group(P0.01 or P0.05).Comparing T group with ANP group, the blood flow of lung in T group was significantly higher than that of ANP group (P0.01),pathological changes were alleviated significantly,lung ratio wet/dry had no change(P0.05),and MPO activity were significantly lower than those in ANP group(P0.01).There were significant negative correlation between the blood flow of lung and dry/wet ratio,MPO activity and the pathological changes of lung in ANP group. CONCLUSION MCD is an important factor of lung injury in ANP. Ligustrazine can ameliorate the condition of MCD and alleviate the damage of pancreas and lung.
