Determination of Esculentoside A in Radix Phytolaccae and Its Dynamic Changes During Steaming Course

Abstract

Objective:To establish a method for the content determination of Esculentoside A in Radix Phytolaccae,and analyze the dynamic changes of Esculentoside A during the course of steaming.Methods:The method of steaming with mung bean was used to process Radix Phytolaccae.HPLC was performed on the SinoChrom ODS-BP column;1%formic acid water-1%formic acid acetonitrile served as mobile phase and the detection wavelength was at 210 nm,the column temperature at 30℃and the flow rate being 1.0 ml/min,to determine the content of Esculentoside A.Results:Esculentoside A showed good linearity in the range of 1.424-7.120μg with correlation coefficient of 0.999 8.The average recovery(n=9) was 99.92%,with RSD of 2.3%. Esculentoside A decreased logarithmically during the course of steaming.Conclusion:This method is simple and shortcut and the result is accurate and reliable.The traditional processing method can indeed reduce the content of toxic composition in Radix Phytolaccae and deserves further studies.
