IFN-γ reinforces the inhibitory effect of tumor necrosis factor-related apoptosis-inducing ligand on apoptosis of human neuroblastoma cell line LAN-1 cells

Abstract

Objectives To explore the influence of IFN-γ on the role of tumor necrosis factor-related apoptosis-inducing ligand(TRAIL) in inducing apoptosis of neuroblastoma LAN-1 cell line.Methods Survival fraction and apop-tosis were measured by MTT method and flow cytometry(FACS).Results Survival fraction of IFN-γ group,TRAIL and IFN-γ 72 h + TRAIL group were(97.12 ± 3.03)%、(83.35 ± 2.95)%、(60.05 ± 4.05)% respectively.The percentage of apoptotic cells of IFN-γ group,TRAIL group,IFN-γ 24 h + TRAIL group,IFN-γ 48 h + TRAIL and IFN-γ 72 h + TRAIL group were(2.26 ± 0.35)%、(3.03 ± 0.68)%、(6.87 ± 0.99)%、(9.80 ± 1.85)%、(15.11 ± 2.57)% respective-ly.The percentage of apoptotic cells of IFN-γ 24 h + TRAIL group was higher than that of IFN-γ group and TRAIL group(P 0.05) .The longer the IFN-γ pretreated,the higher the percentage of apoptotic cells was observed(P 0.05) .Conclu-sions IFN-γ can enforce the effect of TRAIL inducing apoptosis in LAN-1.
