The Study on the Neuroprotective Effects of Resveratrol against Degeneration of Dopaminergic Neurons in Parkinson′s Disease Rat Model

Abstract

Objective To observe the neuroprotective effects of resveratrol(Res) against degeneration of dopaminergic neurons which caused by LPS in vivo.Methods PD rat model were made by intranigral injection of LPS.Sprague-Dawley rats were randomly divided into control group,PD group and Res group.Behaviour study,tyrosine hydroxylase(TH) and nuclear factor kappa B(NF-κB),immuno-histochemistry and Western blot were performed.Results The control group showed no rotational behavior,the number of circling behavior in PD group was the most in all groups(196.90±9.52).there was significant differences compared with control group and Res group(P 0.01).The number of TH positive cells and the expression of TH protein in substantial nigra of PD group significantly decreased compared with control group(P 0.01);the NF-κB positive cells and NF-κB protein expression were upregulated and showed the most of microglias activation.But compared with PD group,the number of TH positive cells and TH protein expression was increased,the NF-κB positive cells and NF-κB protein expression was decreased in Res group(P 0.01).Conclusion Res showed neuroprotective effects against the degeneration of dopaminergic neurons caused by LPS in vivo.
