Abstract
Objective:To study effects of valsartan on ventricular hypertrophy and expression of connective tissue growth factor in the rats with renovascular hypertension (RVH). Method:One group of two-kidney one-clip (2K1C) RVH rats received valsartan (30 mg·kg-1·d-1) and the other group with no treatment for 4 or 8 weeks. The left ventricular weight(LVW) and the ratio of left ventricle weight to body weight(LVW/BW) were measured. The CTGF mRNA and protein in the left ventricular myocardium were assayed by semi-quantitative RT-PCR and immunohistochemical methods.Result:The expression of CTGF mRNA and protein in the left ventricular myocardium increased significantly in RVH rats without treatment but decreased significantly after 4wks or 8wks treatment with valsartan(P0.05). Conclusion:Increased expression of CTGF mRNA and protein in left ventricular myocardium of rats with renovascular hypertension can be partially prevented by valsartan. This may be one of the mechanisms for AT1 receptor antagonists to prevent and reverse left ventricle remodeling.
