Abstract
Objective To investigate effects of amiodarone on potassium channel gene expression in myocardial sleeves of pulmonary vein in rabbits with rapid atrial pacing.Methods Rabbits were divided into control groups(n=10),rapid atrial pacing(RAP) group(RAP group,n=10) and RAP and amiodarone group(Am group,n=10).Myocardium tissues were obtained from rabbit pulmonary veins,the messenger ribonucleic acid(mRNA) of Kir3.1,KvLQT1,HERG of specimen was measured by reverse transcription-polymerase chain reaction.Results Compared with the control group,Kir3.1 mRNA expression reduced by 42.8% in RAP group(P0.01),and 46.4% in Am group(P0.01).The mRNA levels of HERG significantly increased by 28.8% in Am group(P0.01) compared with the control group.There was no significant difference in mRNA levels of KvLQT1 among the three groups.Conclusion The RAP results in changes of the potassium channel gene expression in myocardial sleeves of pulmonary vein in rabbits and amiodarone also has some effect on them.
