Intervention Effect of Cyclosporine A in Vitro on Hematopoietic Cells in Patients with Aplastic Anemia

Abstract

Objective:To observe the effect of cyclosporine A (CsA) in vitro on colony-forming unit-granulocyte macrophage (CFU-GM) and colony-forming unit-erythroid (CFU-E) in patients with aplastic anemia (AA),and to offer experimental evidence to potentialy clinical use of CsA.Methods:Bone marrow mononuclear cells from 54 AA patients were collected and cultured in RPMI 1640 agar and McCoy's 5A mythycellulose medium,respectively.The effects of CsA on CFU-GM and CFU-E from AA patients were compared with those from 30 normal controls.Results:①The numbers of CFU-GM and CFU-E in patients with AA were significantly decreased than those of normal controls (P0.01).②CsA could significantly promote the proliferation of CFU-GM and CFU-E in the patients,and this proliferative effect was dependent on the dose of CsA.③The response rate in severe AA patients to CsA was higher than that in chronic AA patients.④The response rate of patients with severe and moderate decreases and with CFU-GM and CFU-E proliferation was higher than that of those patients without proliferation of CFU-GM and CFU-E.Conclusions:CsA could promote in vitro the proliferation of CFU-GM and CFU-E in some of the patients with AA,therefore,it could be regarded as an effective treatment of AA.
