Study on the anther culture in vitro of cassava

To study the effect of plant growth regulator,sugar concentration,medium and Low temperature pretreatment on cassava anther culture,the anthers of 3cassava varieties were used as explants.The results showed that callus induction frequency was significantly different among 2,4-D,Picloram and TDZ.2,4-D was better than TDZ.The best induction rate was obtained when 2,4-D and 6-BA were co used.Sugar concentration had certain effect on anther culture.While the concentration of sugar exceeded 12%,the anther callus induction was inhibited.Bud through 3days of 4℃low temperature pretreatment could improve the induction rate,but was reduced after 7days,the induction rate was reduced to 0after 21days.The anther callus of Cassava variety GR911and wild cassava just could be inducted to generate in medium adding 6-BA、KT and TDZ.Little GR891anther callus could be induced to grow adventitious roots in the medium adding 6-BA but only propagate callus while adding KT and TDZ without adventitious roots.The most suitable medium of adventitious roots was N6+1.0mg.L-1 6-BA+0.25mg.L-1 NAA+0.5mg.L-1 CuSO4.
