Effect of PAF antagonist BN50739 on acute lung injury in rats

Abstract

Objective To observe the therapeutic effect of BN50739, a platelet activating factor (PAF) antagonist on the acute lung injury induced by burn injury combined with endotoxemia. Methods Third degree thermal injury of 20% total body surface area (TBSA) rats were prepared, and intraperitoneal injection of endotoxin were given to induce animal model of acute lung injury. BN50739 at 20 mg/kg was injected into the abdominal cavity of rats in treatment group 5 min before the injury. Histological changes of the lung, blood gas analyses, protein contents in the bronchoalveolar lavage fluid (BALF) and expression of TNG in lung tissue were investigated before. Results BN50739 significantly attenuated the pulmonary edema, and decreased lung vascular permeability for protein, and resulted in the decrease of the protein contents in BALF improvement of respiration and increase of the blood PO 2. BN50739 partially inhibited the expression of TNF in rat lung. Conclusion BN50739 might partially improve the acute lung injury induced by burns injury combined with endotoxemia in rat.
