Effect of platelet-activating factor acetylhydrolase on severe acute pancreatitis-induced lung injury in rats

Abstract

Objective To investigate the effect of platelet-activating factor acetylhydrolase (PAF-AH) on severe acute pancreatitis (SAP)-induced lung injury in rats.Methods Forty-five male Wistar rats were randomly divided into three groups (n=15 in each group):sham operation group,SAP group and PAF-AH group.PAF-AH (5 mg/kg) was infused by dorsal artery of penis 1 h and 24 h after establishing the model in PAF-AH group.After establishing the animal model for 48 h,the contents of platelet-activating factor (PAF),endotoxin,tumor necrosis factor (TNF-α) in serum and the activity of myeloperoxidase (MPO) in lung tissue were examined.The lung tissue was scored on the basis of pathological changes.Results Compared with SAP group,the contents of PAF (7.91± 0.12) pg/L,endotoxin (0.28± 0.02) EU/ml,TNF-α (1.46± 0.76) μg/L in serum,MPO activity in lung tissue (5.24± 1.34) U/g and the pathological scores of lung (4.88± 1.35) were significantly lowered in PAF-AH group (P 0.05).Conclusion Administration of PAF-AH can reduce the injury of lung by ameliorating the microcirculatory disorders,protecting the intestinal barrier function and inhibiting the acute inflammatory reaction.
