Abstract
Objective Establishing a mouse model of cisplatin ototoxicity,studying the effects of cisplatin of different doses on the expression of Calpain in mouse cochlea.Methods: 70 BALB/c mice were randomly divided into five groups,after intervention with different doses of cisplatin,detecting the auditory changes before and after treatment through the auditory brainstem response tests.Western blot and immunofluorescence were applied to detect the expression of Calpain in the cochlea.Results: The weight and hearing of mice decreased significantly in the goups which treated with different doses of cisplatin.With prolongation of drug administration time,the expression of Calpain1 in the cochlea decreased gradually,while the expression of Calpain 2 in the cochlea increased gradually.Conclusion: Application of cisplatin can establish the mice ototoxicity model.Normal mouse cochlea exists the expression of calpain1 and calpain2,moreover,calpain2 is associated with cisplatin-induced ototoxicity.
