Abstract
Objective To explore the expression of calpain in aging rat cochleas and correlation between change of calpain-calpastatin and age-related hearing loss.Methods Immunohistochemical staining were used to detect the expression and location of calpain in aging rat cochleas,and real time PCR assay were used to detect mRNA level of calpain and calpastatin in cochlear extracts.Results Calpain distributed in outer and inner hair cells,spiral ganglion or stria vascularis.Lower expressions were seen in younger rat cochlear tissues.Calpain expression significantly increased in aging rats than those in younger rats in hair cells,spiral ganglion and stria vascularis.There was a similar expressing pattern in the aging rats with different auditory brain response(ABR) threshold.mRNA level of calpain increased 5.7-fold in aging rats cochleas than that in younger rats.In contrast,mRNA level of calpastatin decreased 3-fold(P0.01).Conclusions The functional disorders of calpain and calpastatin could play an important role in cochlear tissue degeneration and hearing lesion by age-related hearing loss.
