Abstract

Objective To observe the effect of Scutellaria barbata polysaccharides(SBP) on inhibiting the proliferation of hepatoma cell line SMMC7721 and to explore its relevant mechanism.Methods SMMC7721 cells were cultured with different concentrations of SBP.The proliferation of SMMC7721 cells was detected by methyl thiazolyl tetrazolium(MTT) assay.The expression of CDK2 and E2F1 mRNA was detected by using real-time polymerase chain reaction(PCR),and the expression of CDK2 and E2F1 protein was detected by using Western blot method.Results SBP could effectively inhibit the proliferation of SMMC7721 cells,and SBP at concentrations of 2.5 and 10 μg·mL-1 had obvious effect on decreasing the mRNA and protein levels of CDK2 and E2F1.Conclusion SBP can inhibit the proliferation of SMMC7721 cells,and its mechanism is possibly related with the decrease of CDK2 and E2F1 expression.
