Abstract

Objective To study inhibition effect of octreotide on the growth of gastric carcinoma cells SGC-7901.Methods The gastric carcinoma cells were treated with octreotide. The effect of growth suppression on cells was evaluated by MTT assay. The effect of octreotide on DNA synthesis of gastric carcinoma cells was evaluated by 3H-thymidine( 3H-TdR)incorporation.Morphologic changes of gastric carcinoma cells were observed under microscope.Cell cycle was detected by flow cytometry(FCM).The activity of telomerase of gastric carcinoma cells was examined by TRAP PCR-ELISA.Results Octreotide obviously inhibited the growth and DNA synthesis of SGC-7901 cells.SGC-7901 cells became round, small, and exfoliating after treated with Octreotide. The FCM analysis showed that the number of gastric carcinoma cell of G 0/G 1 phase increased and the number of cell of S phase and G 2/M phase decreased. The activity of telomerase of gastric carcinoma cells was obviously inhibited. Conclusion Octreotide can obviously inhibit the growth of SGC-7901 cells, cause G 0/G 1 blocking and inhibit the activity of telomerase.
