Abstract
Objective To study the feasibility of inducing the adipose-derived stem cells into osteoblast cells in vitro with platelet-rich plasma(PRP).Methods Rabbit adipose-derived stem cells were in vitro isolated and cultured into the third passage,then they were assigned into two groups: the control group were treated with 10 mmol/L β-sodium glycerophosphate,50 mg/L ascorbic acid,1×10-8 mol/L dexamethasone.The experimental group were given 10ml/L PRP in addition to all the treatment in the control group.Cell morphological changes and alkaline phosphatase(ALPase) activity were detected by immunohistochemical staining;formation of the mineralzation nodes was detected by von Kossa staining and Alizarin bordeaux staining.Results Adipose-derived stem cells in the experimental group became a regular polygon form.Cell doubling time was significantly shortered as compared to the control group.Over time,alkaline phosphatase expression was increased in the experimental group,and significantly higher than that of control group after 7,10 and 14 days(P0.01).after 21 days,the mineralzation nodes became more and bigger in the experimental group as compared to the control group.Conclusion PRP can effectively induce the rabbit adipose derived stem cells into osteoblasts.
