Abstract
Objective To observe the effect of induction time selection on the differentiation of rat adipose derived stromal cells(ADSCs) into osteoblasts.Methods The induced group was added into conditioned medium containing 1×10-8mol/L dexamethasone,10mmol/L beta-sodium glycerophosphate and 50mg/L ascorbic acid at the same time of seeding,at 50%-60% confluence,at 85%-90% confluence,respectively.The A,B,C group of non-induction group was cultured with ordinary medium.Results After calcium-cobalt staining,in induced group a majority of cells were positively stained,and the quantity of positive cells reached the peak at 85%-95% confluence.Non-induced group was no positive cells;Alizarin red stain were visible red nodules,The induction group formed nodules earlier at cells of 85%-95% confluence;Intracellular alkaline phosphatase activity detection showed that intracellular alkaline phosphatase activity markedly increased after inducing by osteogenic inducer than non-induced group.Conclusion After induced ADSCs can differentiate into osteoblasts;Induced alkaline phosphatase activity within cells was significantly higher than non-induced group;Under the same initial seeding density,the higher the ADSCs contact with each other,the stronger the osteogenesis is.
