Abstract

Obejective To investigate the mechanism of hepatic I/R injury in rats.Methods Thirty-two healthy male SD rats were randomly divided into 4 groups:group A:sham-operated control group,group B:total hepatic ischemia 40min,group C:2h reperfusion after 40min ischemia,and group D:4h reperfusion after 40min ischemia.The histologic sections of liver were made in 4 groups,plasma TNF-α,ALT,LDH,content of ET-1 in liver tissue were respectively examined in the 4 groups.The expression of P-selectin in liver tissue was located and analyzed by immunohistochemical staining.Results Plasma ALT,LDH,TNF-α,as evidenced by significant increase in groups C,D.The content of MDA,an index of lipid peroxidation in liver tissue,went up to(54.16±3.38)μmol/g protein in group C.ET-1 in liver increased significantly in groups C,D.P-selectin in liver expressed highly significantly in groups C,D,as compared with groups A,B.There was histologic evidence of liver injury.Conclusion Activation of endothelin,cytokines and oxygen radicals contribute to microcirculatory disturbance,which is pathologic procedure of hepatic ischemia and reperfusion.
