Experimental study on apoptosis of osteosarcoma cells by recombinant human interferon-α combined with cisplatin

Abstract

Objective:To investigate the effect of combined recombinant human interferon-α(rhIFN-α) with cisplatin on the apoptosis of osteosarcoma cells(MG-63).Methods:We treated the osteosarcoma cells by combining rhIFN-α with cisplatin and the morphological change of the cells were observed by light microscope.The proliferation and apoptosis of osteosarcoma cells were examined respectively by MTT and flow cytometry,and further detection of the apoptosis was carried out with hoechest 33258 staining and DNA ladder solution.Results:Induction by 10 000 IU/ml rhIFN-α could produce evident effect on the ratio of apoptosis(34.12±2.11)% as compared with by single use of cisplatin(24.74±2.22)%,and yet,combined use of rhIFN-α with cisplatin could boost the apoptosis ratio of MG-63 to(69.41±1.72)%,(58.81±2.23)% and(48.23±1.12)%,respectively.Conclusion:The apoptosis ratio of osteosarcoma cells(MG-63) induced by rhIFN-α demonstrated dose dependent.10 000 IU/ml rhIFN-α can significantly induce the apoptosis of MG-63 and different dose of rhIFN-α appears to boost the apoptosis induced by cisplatin,which suggests that combined therapy of cisplatin with rhIFN-α can improve the response of the tumor cells to the medication.
