Abstract
Chitinase-producing Serratia marcescens culture medium components and culture conditions of enzyme production by shake flask fermentation were optimized with uniform-design and orthogonal-design respectively.The results showed that the optimal component concentrations(m/m) were colloid chitin 0.504%,yeast extract 1.178%,MgSO4·7H2O 0.025%,K2HPO4 0.095%,FeSO4·7H2O 0.001%,sorbitol 0.005%,KH2PO4 0.030%.The optimal culture conditions were incubation time 48 h,temperature 30 ℃,initial pH 9.0,loadage 30 mL per 250 mL flask,inoculation quantity 1%.The activity of chitinase reached 9.39 μkat·L-1 which was increased by 81.6% than that of the unoptimized conditions..
