Abstract
Objective To develop a productive,inexpensive and stable method for the isolation and culture of rat hepatic stellate cells (HSCs) as a cell model for the studies of hepatic fibrosis.Methods Rat HSCs were isolated by liver perfusion with pronase and collagenase followed by density gradient centrifugation with Nycodenz.Results The harvest rate of hepatic stellate cells was (3.2±0.67) × 10~7 cells per liver and viability rate was more than 90%.Conclusion This method is an ideal option for the isolation and culture of rat HSCs.
