Application of lymphocyte subgroup detection by flow cytometry in monitoring cellular immune dysfunction with diabetes

Abstract

Objective To detect peripheral blood lymphocyte subsets distribution of diabetes mellitus(DM)patients and get the message about the changes of cellular immune function in DM.Methods The levels of T cells(CD3+,CD3+CD4+and CD3+CD8+),B cells(CD3+CD19+),NK cells(CD3+CD16-56+),and activated T cells(CD3+HLA-DR+and CD3+CD25+)were detedted by Flow cytometry with three-color fluorescence labeling of monoclonal antibodies in 459 cases of healthy persons and 100 cases of DM in patients.Cell percentage from 98% confidence interval,P0.05 with statistical significance.Results DM patients with CD3+,CD3+CD8+cells percentage were higher than that of the control group(P0.01).DM patients with CD3+CD19+,CD3+HLA-DR+,CD3+CD25+cells percentage were lower than the control group(P0.01).There were no significant differences in T4/T8 ratio and the percentage of NK cells between DM patients and health persons.Conclusion There is a cellular immune dysfunction in diabetics patients.Lymphocyte subsets detection can be used as indicators of clinical diagnosis of diabetes.
