Cloning and Sequence Analysis of Human Interleukin-38 Gene

Abstract

Objective: To clone and analyze the cDNA sequence encoding human interleukin(IL)-38 gene.Method:Total RNA was prepared from the skin of human,cDNA fragment encoding IL-38 gene was amplified by RT-PCR using specific primers,and then was cloned into pcDNA3.1 vector.Recombinant eukaryotic expression vector pcDNA3.1-hIL-38 was constructed and was transformed into the host E.coli strain TOP10 for identification.Inserted IL-38 gene was sequenced.Result: Sequence analysis indicated that the cDNA of IL-38 had the whole length of 459 bp with a complete open reading frame,which encodes the product of 152 amino acids.The cDNA shared 100% homology with the sequence of mRNA for human IL-38 gene in GenBank.Conclusion: The whole-length cDNA of human IL-38 was successfully cloned,which laid a foundation for further studying on its biological function.
