Abstract
Objective: To investigate the transfection efficiency and cell toxicity of Polyethyleneimine (PEI) derivative linking with 1, 4-Dibutanediol Chloroformate. Methods: Luciferase plasmid was used as the reporter gene to investigate transfection efficiency in Hep G2 cells, cytotoxicity experiments were carried out in this cell line either. Results: Transfection results indicated that the gene carrier (PEI-Bu) performed better than the positive control of PEI 25kDa and Lipofectamine 2000. Cytotoxicity result suggested that PEI-Bu was less cytotoxic than PEI 25kDa with the increase of polymer concentration. Conclusion: Transfection and cytotoxicity data of Hep G2 indicated that PEI-Bu could be a highly efficient and less toxic gene carrier, which was promising in liver gene therapy.
